چکیده

Dyslipidemia has been associated with premature atherosclerosis and other pathological conditions, and the determination of serum lipids is of great importance in health and disease. Most of the lipids are transported in plasma in the form of lipoproteins. Four classes of lipoproteins have been recognized by electrophoresis: chylomicrons, beta-, pre-beta-, and alpha-lipoproteins. Fredrickson, Levy, and Lees (1) classified the hyperlipoproteinemias into five types by paper electrophoresis; their classification is a very useful diagnostic and therapeuticaid. The renewed interest in serum lipoproteins stimulated the development of improved electrophoretic methods for their determination (2-5).

Abrnomal lipoprotein patterns in human serum as determined by agarose gel electrophoresis.

Two groups of abnormal electrophoretic patterns of serum lipoproteins are reported here. One demonstrates a deficiency or absence of lipoprotein fractions, which is characteristic of patients with abeta-lipoproteinemia, hypo-beta-lipoproteinemia, or Tangier disease. The other shows the presence of extra lipoprotein fractions, as found in cholestasis and multiple myeloma. These patterns, togethe...

Quantitative measurement of lipoprotein surface charge by agarose gel electrophoresis.

The electrophoretic mobilities of low density lipoprotein (LDL) and six pure proteins in a 0.5% agarose gel have been compared to literature electrophoretic mobility values determined by the Tiselius moving boundary method. There is a strong correlation (r = 0.99) between the electrophoretic mobilities determined by the two techniques. The electrophoretic behavior of charged particles smaller t...

Evaluation of light-scattering index (nephelometry) for assessing serum triglycerides and lipoprotein phenotypes.

Light-scattering measurements (nephelometry) of serum after filtration through appropriate filters is a rapid method for physically determining triglyceride concentrations and lipoprotein phenotypes. A correlation coefficient of greater than 0.9 was obtained when aliquots of the same specimen were tested for triglycerides by light scattering in an Aminco-Bowman Spectrophotofluorometer and by a ...

Quantitation of serum lipoproteins by electrophoresis on agarose gel: standardization in lipoprotein concentration units (mg-100 ml) by comparison with analytical ultracentrifugation.

Lipoprotein electrophoresis on agarose gel has been modified to allow estimation of the absolute quantity of each fraction. The reproducibility of the method is illustrated by 12 determinations in a single day on serum from one normal subject: mean total dye uptake was 302 +/- 9 (sd "corrected dye units," and the percentages of beta-, pre-beta, and alpha-lipoprotein were 56.1 +/- 0.9, 29.1 +/- ...

Microscope slide electrophoresis of serum lipoproteins in agarose gel.

An ultramicro electrophoretic technique for the separation of serum lipoproteins is described using agarose gel on microscope slides. The interaction between the agarose gel and lipoproteins has been reduced and the separation between beta- and pre-beta-lipoproteins has been improved over existing methods. A semiquantitative assay of the lipoproteins can be done in a densitometer. It is suggeste...
